ANTIOXIDANT ACTION OF THE FULL SPECTRUM CANNABIDIOL OIL ON DYSTROPHIC MUSCLE OF MDX MICE

Introdução

Oxidative stress in Duchenne muscular dystrophy (DMD) can strongly contribute to the progression of the disease, leading to the exhaustion of myogenic cell capacity and potentiating the process of cellular damage. Cannabidiol (CBD) exerts several beneficial effects, including anti-inflammatory and antioxidant properties.

Objetivo

The objective is to evaluate the antioxidant action of full-spectrum CBD in dystrophic mice.

Método

Approved by the Ethics Committee on the Use of Animals, we assessed wild-type mice (C57BL/10; WT) and mdx mice for the in vivo analysis, dividing them into three treatment groups: saline (mdxCT-; n = 7), grape seed oil (mdxCT+; n = 7), and full-spectrum CBD oil at 10 mg/kg body weight for 14 days (mdxCBD; n = 7). Twenty-four hours after the last full-spectrum CBD or vehicle treatment, the animals were anesthetized, perfused, and the quadriceps muscle (QUA) and diaphragm (DIA) were collected for analysis of the levels of the antioxidant enzyme catalase (CAT), glutathione (GSH), 4-Hydroxy-2-Nonenal (4-HNE), and the presence of lipofuscin granules. The group means were compared using analysis of variance (ANOVA) followed by the Tukey test for multiple statistical comparisons between groups.

Resultados

The mdxCBD group was able to increase CAT levels in QUA (67%) and DIA (62%), increase GSH in QUA (70.1%), and reduce the number of lipofuscin granules in QUA (41%) and DIA (82%) compared to the mdxCT- group. However, there was no significant difference in the analysis of 4-HNE levels.

Conclusão

In this study, we identified that full-spectrum CBD has positive effects on oxidative stress in the DMD model.
